To join the ranks of the Sisterhood as an Acolyte go to Wallach, venture 4w, then 'enter' the Headquarters.

Regen points are earned by increasing your stats, specifically Wis, Con, and Qui.
Every guild skill (active or passive, combat or noncombat) gives some amount of guild experience (gexp). You use this gexp to improve your gskills and guild levels.

Gskills can be trained up to 100% in the room 2w of the main guild room. Gskills cost gexp and solaris to improve.
Be aware that when you level up your guild levels, you won't obtain skills immediatly, they take a little while to register. You will get a message saying 'You have learned the x ability.' when it is ready. Once you get this message, you can go train up in that skill.
Guild level gives you passive skills that either buffs your stats, raises defences/offenses or give stat bonuses.

When you get your first literjon (the blue one) you can fill it up with spice at the spice bar in the guild. Once you have unlocked this, you will need to start keeping your spice levels up or your skills will suffer higher chance of failure (amongst other things i'm sure but what they are, i don't know!)
Few useful commands:
Your monitor will show you your spice saturation level, be careful about going above 100% because spice comas are real and once it happens you're stuck lying on the floor helpless until it abates.
Here's a real quick set of tips to make the low-level BG experience less awful. The goal is to maximize your gexp rate as much as possible.

At this point you should be doing OK gexp-wise. Short term goals at this point include maxing body-hardening, palmstrike, agony, and prana-bindu; getting Bindu Level 10 and training bandage
; evening out your stats, Wis, Qui, Dex especially; and training mud skills like Hand to Hand, Dodge, and Block.
Medium term goals should include unlocking and training counters (glvl 45), weirding and blitz (plvls 20 and 40), and rupture (mlvl 40). At Guild Level 50 you get to start managing your spice-level.
Here's some thoughts about the four trees (glvl, prana, bindu, manip) of skills.
Endgame, your priority of training should be prana > guild level » manipulation. There's very little reason to raise Bindu above 100.
